The seeds of all plants are subject to intense bursts of oxidative stress as they undergo the biological process of maturation, and as such they have evolved a very complex and effective antioxidant system. Many of the key components of the antioxidant system are mediated by enzymes. Soybeans are a convenient source of biomass for the isolation of components of the antioxidant system such as phytomaterials due to their ready supply. Soybean Peroxidase is a 37 kD protein typically isolated from the seed coat that catalyzes the conversion of Hydrogen Peroxide (H2O2) to water.

Hydrogen Peroxide is a very useful molecule in moderation. While the industrial applications for peroxide are rapidly growing, the cosmetic uses remain limited with the focus being on hair care products. However, there are numerous ways in which peroxide can enter into a cosmetic product other than direct addition. Perhaps, the most common reason for the presence of peroxide is through the inclusion of other enzymes in a formulation. These inclusions could be for a variety of reasons, but most commonly as an antimicrobial, a whitening or dyeing product, or a general oxidative agent where required.


SCIENCE

FSS Ox-Red Soy Peroxidase PF has been standardized to greater than 200 μM/ml. The associated chaperone proteins (responsible for reacting with the protein ie. bonding or folding to facilitate assembly/disassembly etc.) isolated with the peroxidase to serve to increase stability in solution. While many oxidases such as Glucose Oxidase will generate hydrogen peroxide, the most common actor is Superoxide Dismutase (SOD) which occurs within the skin and body naturally but is additionally commonly added as an active ingredient in many cosmetic formulations. SOD catalyzes the neutralization of the Superoxide Anion to Hydrogen Peroxide. While there are clearly benefits to the approach of neutralizing these peroxides, unless steps are taken to remove the peroxide molecules, this merely moves the oxidative stress from an aqueous environment to a lipophilic one. This occurs because the peroxide reaction in non-lipid molecules is non-propagating, meaning it doesn’t cascade. Once an oxidant is neutralized, that’s the end of the reaction. In lipids, the process continues and effects all the surrounding lipid molecules. The potential damage to skin or hair by peroxide is significantly increased by the presence of Iron. Hydrogen peroxide in the presence of ferrous iron (Fe2+) will generate the hydroxyl radical (OH.) which has an even higher oxidation potential than hydrogen peroxide itself. Under proper conditions, this process can be catalytic with the ferrous iron being regenerated. This reaction developed by H. Fenton in the late 1800’s is still used as the basis for some water treatment applications.


 It is possible through the use of antioxidants to significantly inhibit the formation of lipid peroxides. The study is conducted in a Lucite filled chamber fitted with sample ports and an oxygen electrode. The chamber is charged with 20 ml of distilled water and oxygen levels are measured to establish baseline. 5.5 grams of a linoleic acid/ polysorbate-20 (10:1 w/w) is added to the chamber along with the test material. 2,2’ azo-bis dihydrochloride (0.5M aq. Soln.) is used to initiate the oxidation reaction.
